This is a working overview of mass spectrometry, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-03-23 and is reviewed periodically as new material appears.
Identity and purity are established by instrumental methods rather than by appearance. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and yields a purity value, usually expressed as the share of total peak area. Mass spectrometry checks that the observed mass agrees with the mass calculated from the published sequence, while peptide mapping or amino acid analysis adds structural evidence. Water content, counter-ion identity and residual solvents are sometimes reported as well. A certificate of analysis should name the method behind each figure, because results are method-dependent.
Laboratory handling follows the conventions used for other synthetic peptides. Lyophilized material is weighed and dissolved in an aqueous diluent, typically sterile water or bacteriostatic water, using gentle swirling rather than vigorous shaking, because foaming stresses the chain. Solutions are prepared under clean conditions and, where sterility matters, passed through a suitable filter. Working portions are kept small so that stock material is not repeatedly warmed and cooled, a practice that limits both aggregation and gradual loss of activity.
Stability depends strongly on physical state. Dry powder is comparatively robust when held at -20 °C or below, desiccated and shielded from light; under those conditions degradation is slow and measured over years. Once dissolved, the peptide becomes far more vulnerable: backbone hydrolysis, oxidation of susceptible residues and aggregation all proceed faster in solution, and the rates climb with temperature and with pH far from neutral. Refrigerated storage at 2–8 °C extends usable life for short periods, and repeated freeze–thaw cycles are best avoided.

Dissolution behavior depends on the amino acid sequence, the counterion content, and the buffer chosen. Many peptides disperse readily in water or mild aqueous buffers, while others require a small amount of organic co-solvent or a change in pH. Adsorption to plastic and glass surfaces can reduce the concentration of a solution over time, particularly at low concentrations. Filtration before analysis removes particulates, and aliquoting limits repeated freeze-thaw cycles that stress the material.
Research-grade peptide material is commonly supplied as a lyophilized powder, a form that limits degradation during transport and storage. Standard practice keeps such material cold and protected from light and moisture, with tighter conditions used for long-term archives. Once dissolved, solutions are generally considered less stable than the dry powder and are handled on shorter timescales. These established conventions derive largely from general peptide chemistry rather than from compound-specific evidence alone.
Identification and purity assessment typically rely on reversed-phase high-performance liquid chromatography, often paired with mass spectrometry. Mass measurement confirms the expected molecular mass and can reveal truncations or modifications. Peptide mapping and sequencing techniques provide sequence-level confirmation when needed. Because related peptide impurities can behave similarly in a single method, orthogonal techniques are usually combined. Reported purity values depend heavily on the method used and should be interpreted with that in mind.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Retatrutide is an investigational synthetic peptide engineered to activate three distinct hormone receptors within a single molecule. It targets the glucose-dependent insulinotropic polypeptide receptor, the glucagon-like peptide-1 receptor, and the glucagon receptor simultaneously. This triagonist design distinguishes it from earlier incretin-based compounds that act on one or two of these pathways. Structural modifications relative to native gut hormones extend its residence time in circulation. The molecule remains under clinical evaluation and is not approved for any indication.
In 2022, Biden blocked a national railroad strike planned by multiple labor unions. During the United Auto Workers strike, he expressed support for the workers. Biden joined striking UAW workers' picket line in Michigan, becoming the first president to join a picket line. He refused to block a port strike from the International Longshoremen's Association in October 2024. Over the course of five days in March 2023, three small- to mid-size U.S. banks failed, triggering a sharp decline in global bank stock prices and swift response by regulators to prevent potential global contagion. After Silicon Valley Bank collapsed, the first to do so, Biden expressed opposition to a bailout by taxpayers. He claimed that the partial rollback of Dodd-Frank regulations contributed to the bank's failure. At the beginning of the 118th Congress, Biden and congressional Republicans engaged in a standoff after the U.S. hit its debt limit, which raised the risk that the U.S. would default on its debt. Biden and House speaker Kevin McCarthy struck a deal to raise the debt limit, the Fiscal Responsibility Act of 2023, which suspended the debt limit until January 2025. Biden signed it on June 3, averting a default. The deal was generally seen as favorable to Biden. Biden extended the COVID-19 student loan pause through September 2023, with an "on ramp" period that extended some of the pause's protections against credit reporting, collection efforts, and late payment fees through September 30, 2024.
== Raids and arrests == In July 2022, the Central Office for the Repression of Corruption (French: Office central pour la répression de la corruption, OCRC, Dutch: Centrale Dienst voor de Bestrijding van Corruptie), a unit of the Belgian Federal Police, opened an investigation into an alleged criminal organisation. The investigation was led by Michel Claise, an investigating magistrate who works with the GRECO (Group of States against Corruption). On 9 December 2022, acting on the investigation, Belgian police conducted 20 raids at 19 different addresses across Brussels in connection with the conspiracy, resulting in eight arrests across Belgium and Italy. The homes and offices of the suspects were searched, including offices within the European Parliament buildings in Brussels. As required by the Belgian Constitution, the President of the European Parliament, Roberta Metsola, had to return from her home in Malta to be present during searches of the properties of Eva Kaili and Marc Tarabella, both of whom who held parliamentary immunity as MEPs. The first arrest took place at Sofitel hotel at Place Jourdan, Brussels, where investigators arrested Eva Kaili's father, Alexandros. He was found with a suitcase containing "several hundred thousand euros" while attempting to flee. Investigating judge Michel Claise deemed the arrest in flagrante delicto, or caught in the act, which resulted in Kaili losing her parliamentary immunity, enabling investigators to search her property.
=== Imaging === Medical imaging may provide a limited amount of information regarding bone marrow. Plain film x-rays pass through soft tissues such as marrow and do not provide visualization, although any changes in the structure of the associated bone may be detected. CT imaging has somewhat better capacity for assessing the marrow cavity of bones, although with low sensitivity and specificity. For example, normal fatty "yellow" marrow in adult long bones is of low density (-30 to -100 Hounsfield units), between subcutaneous fat and soft tissue. Tissue with increased cellular composition, such as normal "red" marrow or cancer cells within the medullary cavity will measure variably higher in density. MRI is more sensitive and specific for assessing bone composition. MRI enables assessment of the average molecular composition of soft tissues and thus provides information regarding the relative fat content of marrow. In adult humans, "yellow" fatty marrow is the dominant tissue in bones, particularly in the (peripheral) appendicular skeleton. Because fat molecules have a high T1-relaxivity, T1-weighted imaging sequences show "yellow" fatty marrow as bright (hyperintense). Furthermore, normal fatty marrow loses signal on fat-saturation sequences, in a similar pattern to subcutaneous fat. When "yellow" fatty marrow becomes replaced by tissue with more cellular composition, this change is apparent as decreased brightness on T1-weighted sequences.
This constriction was found to be competitively antagonized by prazosin (a standard α1 antagonist) and ketanserin, with prazosin being the more potent antagonist (pA2 = 9.38, vs pA2 = 8.23 for ketanserin). Synephrine constrictions were also antagonized by BRL-15,572, but not by SB-216,641 (used here as a selective 5-HT1B antagonist), or by propranolol (a common β antagonist). In studies on guinea pig atria and trachea, Jordan and co-workers also found that synephrine had negligible activity on β1 and β2 receptors, being about 40000x less potent than norepinephrine. Experiments with cultured white fat cells from several animal species, including human, by Carpéné and co-workers showed that racemic synephrine produced lipolytic effects, but only at high concentrations (0.1-1 mM). The potency, expressed in terms of pD2 of synephrine in these species was as follows: rat: 4.38; hamster: 5.32; guinea pig: 4.31; human: 4.94. In comparison, isoprenaline had a pD2 = 8.29 and norepinephrine had pD2 = 6.80 in human white fat cells. The lipolytic effect of 1 mM/L of synephrine on rat white fat cells was antagonized by various β-antagonists with the following inhibitory concentrations (IC50): bupranolol: 0.11 μM; CGP-20,712A (β1 antagonist): 6.09 μM; ICI-118,551 (β2 antagonist): 3.58 μM; SR-5923A (β3 antagonist): 17 μM. The binding of racemic synephrine to cloned human adrenergic receptors has been examined: Ma and co-workers found that synephrine bound to α1A, α2A and α2C with low affinity (pKi = 4.11 for α1A; 4.44 for α2A; 4.61 for α2C).
==== Princes of the Royal Secret ==== In April 1818, a member of the Grand Orient de France named Louis de Clouet, who went by the Masonic codename d'Obernay, established a Grand Consistory of the Scottish Rite in Havana, Principes del Real Secreto (English: Princes of the Royal Secret). D'Obernay authorized the Grand Consistory to establish Lodges, chapters, and councils. He also authorized it to confer the 32nd Degree. The Grand Orient of France further authorized this work under the charter of the Grand Consistory dated April 7, 1819. Exactly how many Lodges were merged to create Principes del Real Secreto is debated. However, D'Obernay and this Grand Consistory did create several lodges, including L'Humanite, and La Constante Sophie, which received its charter directly from the Grand Orient of France. The Blue Lodges of the Grand Consistory were formed under the Gran Oriente Simbolico de la Nueva Thebaida (English: Symbolic Grand Orient of New Thebes), which was also known as the Gran Oriente Simbolico de la Isla de Cuba (English: Symbolic Grand Orient of the Island of Cuba). When the Grand Consistory was established, the Grand Orient of France relinquished their authority, handing it over to the National Grand Oriente of Spain. However, because the Grand Orient of Spain was bifurcated politically, the Cubans rejected its authority, preferring to remain autochthonous, instead of finding themselves on either side of the mainland Spanish divisions.

=== Infection === If a chronic wound becomes more painful this is a good indication that it is infected. A lack of pain however does not mean that it is not infected. Other methods of determination are less effective.
==== Mechanism of action ==== Cannabinoid inhibits the activity of both cannabinoid receptor 1 (CBR1) and cannabinoid receptor 2 (CBR2), with a stronger inhibitory effect observed in the former. Cannabidiol also interacts with non-cannabinoid receptors, including serotonin 1A receptors. Yet, the mechanism of cannabinoid in controlling seizures is not clearly established.
== Diagnosis == Since arteritic AION is similar in presentation to non-arteritic AION, patients over the age of 50 diagnosed with NAION must be evaluated to exclude AAION (symptoms: painful jaw muscle spasms, scalp tenderness, unintentional weight loss, fatigue, myalgias and loss of appetite); NAION patients over the age of 75 should always be tested. The distinction between AAION and non-arteritic AION was made to highlight the different etiologies of anterior ischemic optic neuropathy. AAION is due to temporal arteritis (also called giant-cell arteritis), an inflammatory disease of medium-sized blood vessels (Chapel-Hill-Conference) that occurs especially with advancing age. In contrast, NAION results from the coincidence of cardiovascular risk factors in a patient with "crowded" optic discs. Non-arteritic AION is more common than AAION and usually occurs in slightly younger persons. While only a few cases of NAION result in near total loss of vision, most cases of AAION result in nearly complete vision loss. Nonarteritic anterior ischemic optic neuropathy is an isolated white-matter stroke of the optic nerve (ON). NAION is the most common cause of sudden optic nerve-related vision loss, affecting more than 10,000 Americans every year, often bilaterally. No clinically effective treatments exist, largely because little is known about its pathophysiology, and there are few histopathological studies of the acute condition.
Supplementary motor area A region of the medial frontal cortex involved in the planning and coordination of complex, internally generated movements. The SMA contributes to motor sequence initiation, bimanual coordination, and motor imagery. Synapse The junction between two neurons (or between a neuron and another cell) where information is transmitted via neurotransmitters or electrical signals. Synaptic cleft The small space between the presynaptic and postsynaptic neurons across which neurotransmitters are released. Synaptic plasticity The ability of synapses to strengthen or weaken over time, based on activity levels. It underlies learning and memory. Synaptogenesis The formation of new synapses between neurons. It occurs extensively during development and continues throughout life in certain brain regions. Synaptopathy A disorder of synaptic function, often implicated in neurodevelopmental and neurodegenerative diseases such as autism and Alzheimer’s disease. Syringomyelia A condition in which a cyst forms within the spinal cord, potentially damaging nerve fibers and leading to pain, weakness, and sensory loss.
== Political career == In the 1980s and 1990s, Findlay was a supporter of the Liberal Party of Canada. She attended national conventions as a delegate, was an executive member of the Liberal Party Women's Commission, and an executive member of the party for the Vancouver Centre riding association. Findlay entered politics during the 2000 federal election as a Canadian Alliance candidate in the riding of Vancouver Quadra, but lost to Liberal candidate Stephen Owen.

=== Dried flour === In South-west Nigeria, unripe plantains are also dried and ground into flour which is referred to as Elubo Ogede. it is considered as a healthy and nutritious food among the Yoruba. In southern India, dried plantain powder is mixed with a little bit of fennel seed powder and boiled in milk or water to make baby food to feed babies until they are one year old.
== Drug derivatives == Melamine is part of the core structure for a number of drugs including almitrine, altretamine, cyromazine, ethylhexyl triazone, iscotrizinol, meladrazine, melarsomine, melarsoprol, tretamine, trinitrotriazine, and others.
M + Ph2CO → M+Ph2CO•− Generally sodium is used as the alkali metal. Sodium-benzophenone ketyl is used in the purification of organic solvents, particularly ethers, because it reacts with water and oxygen to give non-volatile products. Adsorbents such as alumina, silica gel, and especially molecular sieves are superior and far safer. The sodium-benzophenone method is common since it gives a visual indication that water, oxygen, and peroxides are absent from the solvent. Large scale purification may be more economical using devices which utilize adsorbents such as the aforementioned alumina or molecular sieves. The ketyl is soluble in the organic solvent being dried, which leads to faster purification. In comparison, sodium is insoluble, and its heterogeneous reaction is much slower. When excess alkali metal is present a second reduction may occur. The twice reduced specie presents as a clear red (extra-dry) solution. A color transformation from deep opaque blue (pure once-reduced benzophenone) to red with stages of opaque purple, indicate a mix of anion and bisanion.
Stumptown Coffee Roasters is a coffee roaster and retailer based in Portland, Oregon, United States. The chain's first location opened in 1999. Three other cafes, a roastery and a tasting annex have since opened in Portland, as well as locations in Seattle, New York, and Pasadena. Stumptown is owned by Peet's Coffee, which in turn is owned by Keurig Dr Pepper. The company was an early innovator with cold brew coffee in nitro cans and have continued to develop other cold brew product innovations.

Purity is normally given as a percentage from reversed-phase HPLC, calculated as the main peak area relative to total peak area. Research-grade material is commonly specified at 95 per cent or higher, with tighter specifications available. The number is method-dependent and should be read alongside the chromatogram.
Mass spectrometry is the standard check, comparing the measured mass with the mass calculated from the published amino acid sequence. Retention time on HPLC and peptide mapping provide supporting evidence. Sequence-level confirmation separates it from closely related analogues.
Dry powder is chemically stable enough for freezer storage over long periods. In solution, water participates directly in hydrolysis and enables aggregation, so breakdown accelerates. Cold, dark, short-term storage after dissolution reflects that difference.
常用质谱测定分子量,再结合肽图或序列分析验证一级结构。单一检测手段一般难以排除结构相近的类似物。多种方法相互印证更为可靠。
