Development has progressed from single- and multiple-ascending-dose studies in healthy volunteers into larger randomized trials in adults with obesity, type 2 diabetes, and fatty liver disease. Early reports describe dose-dependent reductions in body weight and improvements in glycemic markers over treatment periods of several months. Whether the glucagon arm adds tolerability cost without added benefit is still debated. Long-term cardiovascular outcomes, effects after treatment stops, and performance in older or comorbid populations are open questions rather than settled findings. Approval status may change, so the current investigational label should be confirmed against regulatory sources.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.
Pharmacologically the compound activates three receptors: GLP-1, GIP, and glucagon. GLP-1 and GIP signaling contribute to glucose-dependent insulin release, delayed gastric emptying, and reduced appetite, while glucagon receptor activation is associated with increased energy expenditure and hepatic fat oxidation. The single-molecule design is intended to keep these activities in one peptide rather than combining separate agents. Relative activity at each receptor differs, and the balance between them is a central question in interpretation. The glucagon component is partly offset by incretin-mediated insulin secretion, an interaction that remains incompletely characterized.

Solid peptide is generally held as a lyophilised powder at low temperature to slow degradation, with desiccant to limit moisture uptake. Reconstituted solutions are less stable and are usually kept refrigerated and protected from light for short periods. Repeated freeze-thaw cycles are avoided because they encourage aggregation. Laboratory handling includes work in a fume hood or laminar flow cabinet to limit inhalation and contamination. Weighing and transfer steps are performed with antistatic tools to reduce static-driven loss of fine powder.
Characterising a peptide of this size relies on a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact molecule from related impurities, while electrospray mass spectrometry confirms molecular mass and detects truncation or oxidation products. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates modified residues. Because the molecule carries a lipid chain, assays must also distinguish the correctly conjugated product from incompletely acylated species.
Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.
Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
As with cocaine in Coca-Cola, lithium was widely marketed as one of several patent medicine products popular in the late 19th and early 20th centuries and was claimed to be included in many drinks including lithia water and 7 Up. Charles Leiper Grigg, who launched his St. Louis-based company The Howdy Corporation, invented a formula for a lemon-lime soft drink in 1920. The product, at one point named "7Up Lithiated Lemon Soda", was launched two weeks before the Wall Street Crash of 1929. It claimed to contain the mood stabilizer lithium citrate, and was one of many patent medicine products popular in the late-19th and early-20th centuries. All references to lithium were removed in 1937 after it became clear that 7Up contains no lithium. Many sources written by scientists (instead of historians) incorrectly report that 7 Up was forced to remove lithium in 1948, with an FDA action that supposedly banned lithium from beverages. Despite the supposed ban, in 1950, the Painesville Telegraph still carried an advertisement for a lithiated lemon beverage.
In August 2004, Palatin signed an agreement with King Pharmaceuticals to co-develop bremelanotide in the US and jointly license it outside the US; King paid Palatin $20 million upfront. Palatin conducted Phase II trials of intranasal bremelanotide in both female sexual dysfunction (FSD) and male erectile dysfunction (ED), but these trials were halted by the FDA in 2007, due to increased blood pressure in clinical trial subjects; Palatin stopped development of the intranasal formulation in 2008. Four trials were conducted in ED, the last being a Phase IIb published in 2008. King terminated the co-development agreement shortly after the FDA halted the trials. The drug was then reformulated to be delivered by injection and trials continued in FSD. A phase II dose-finding trial in FSD in which the drug was administered 45 minutes before sex showed promise at the highest dose and only transient signs of high blood pressure; two Phase III trials were launched at the end of 2014. Palatin launched the Phase III trials with bremelanotide administered via an autoinjector. In 2014, Palatin licensed European rights to bremelanotide to Gedeon Richter Plc. for around $10 million, and Palatin received a milestone payment of around $3 million when it started the Phase III trials in the US. In September 2016, Palatin and Gedeon Richter terminated that agreement. In November 2016, Palatin announced results of the Phase III trials, and shortly thereafter began seeking a partner to complete development in the US.

=== Monoamine oxidase inhibitors === It is unclear whether mescaline is metabolized by monoamine oxidase (MAO) enzymes or whether monoamine oxidase inhibitors (MAOIs) might increase the effects of mescaline. No clinical studies of mescaline in combination with MAOIs are known to have been published. However, there are preliminary reports that harmala alkaloids, which are reversible inhibitors of monoamine oxidase A (RIMAs), may potentiate the effects of mescaline in humans, and the combination of mescaline or mescaline-containing cacti with harmala alkaloids has been referred to as "peyohuasca". In accordance with these findings, the harmala alkaloid and RIMA harmine has been reported to augment the effects of mescaline in animals. On the other hand, Daniel Trachsel has stated that mescaline is not metabolized by MAO, instead being metabolized by semicarbazide-sensitive amine oxidase (SSAO), and hence that MAOIs that do not also inhibit SSAO should have no effect on the metabolism of mescaline. Accordingly, the MAOI iproniazid did not potentiate the behavioral effects of mescaline in rats.
== Development and social impact == John Rolfe of Virginia used this species of tobacco to break the Spanish monopoly on tobacco in 1612. He did so by obtaining Nicotiana tabacum - the coveted plant he had designs to replace Nicotiana rustica with. Up until 1612, Nicotiana rustica was Virginia's only tobacco crop, and was found by Englishmen to be too bitter for enjoyment. Rolfe, by successfully planting, maintaining, and curing Nicotiana tabacum until sale (by removing all superfluous leaves and applying as little water as possible during curing), made Virginia a potential competitor with Spain's colonies. This transitioned the colony to a cash-crop focused one, monocropping tobacco by 1613, the year the first shipment (of 4 barrels) arrived in London. By 1617, the quality of Virginia tobacco had made it the gold standard in European markets, which have become insatiable- 20,000lbs of tobacco was exported there that year. These exports grew exponentially, doubling to 40,000lbs the following year (the year the first enslaved Africans arrive in America), and reaching 60,000lbs by 1620. The money obtained from tobacco exports was used, in turn, to fund the purchasing of more European indentured servants and especially enslaved Africans to fulfill the rising labor demand. In the 17th century, 75% of all colonists in Virginia had come as servants, indentured (with a term of 4 to 7 years of service) or Africans in perpetuity (for life), the latter being preferred as they were cheaper sources of labor:
== Target and events == The primary target of the doctrine was originally the largest communist country, the Soviet Union. However, additional targets also included the other communist countries like the China, Laos, Vietnam, and North Korea. After the dissolution of the Soviet Union in 1991 and China's economic prominence in the early 21st century, China became the major target of the doctrine.
=== Political === When the Allies achieved victory, Mexico was among the victors, despite only actively sending soldiers in the last year of the conflict. Therefore, the country was a founding member of the United Nations Organization, unlike the founding of the League of Nations in 1919, where it was not invited because it had remained neutral during the First World War. In the international arena, Mexico was more present, taking part in the Dumbarton Oaks Conference, the Treaty of San Francisco, the Bretton Woods Conference, and managed to have its initiative approved so that the dictatorship of Francisco Franco in Spain would not be recognized or admitted as a legitimate government before the UN, because it had been formed with the military aid of Nazi Germany and Fascist Italy.
Desirable: <14 mg/dL (<35 nmol/L) Borderline risk: 14–30 mg/dL (35–75 nmol/L) High risk: 31–50 mg/dL (75–125 nmol/L) Very high risk: >50 mg/dL (>125 nmol/L) Lp(a) appears with different isoforms (per kringle repeats) of apolipoprotein; 40% of the variation in Lp(a) levels when measured in mg/dl can be attributed to different isoforms. Lighter Lp(a) are also associated with disease. Thus, a test with simple quantitative results may not provide a complete assessment of risk. The US FDA has given the Tina-quant® lipoprotein Lp(a) RxDx assay from Roche a Breakthrough Device Designation. The assay is designed to identify patients who may benefit from therapies aimed at decreasing Lp(a) levels.

Ion mobility spectrometry–mass spectrometry (IMS-MS) is an analytical chemistry method that separates gas phase ions based on their interaction with a collision gas and their masses. In the first step, the ions are separated according to their mobility through a buffer gas on a millisecond timescale using an ion mobility spectrometer. The separated ions are then introduced into a mass analyzer in a second step where their mass-to-charge ratios can be determined on a microsecond timescale. The effective separation of analytes achieved with this method makes it widely applicable in the analysis of complex samples such as in proteomics and metabolomics.
Epitiostanol, also known as 2α,3α-epithio-4,5α-dihydrotestosterone (2α,3α-epithio-DHT) or as 2α,3α-epithio-5α-androstan-17β-ol, is a synthetic androstane steroid and a derivative of DHT. Mepitiostane, a derivative of epitiostanol with a C17α methoxycyclopentane ether substitution, is an orally active prodrug of epitiostanol. Another derivative, methylepitiostanol (2α,3α-epithio-17α-methyl-5α-androstan-17β-ol), has a methyl group at the C17α position and is similarly an orally active variant of epitiostanol; it has surfaced as a novel designer steroid.
=== Sex differences === There are sex-dependent differences in regional fat distribution. Males are more susceptible to upper-body fat accumulation, most likely in the belly, due to sex hormone differences. When comparing the body fat of men and women it is seen that men have close to twice the visceral fat as that of pre-menopausal women. In women, estrogen is believed to cause fat to be stored in the buttocks, thighs, and hips. When women reach menopause and the estrogen produced by ovaries declines, fat at their buttocks, hips, and thighs decreases while fat at their belly increases. 50% of men and 70% of women in the United States between the ages of 50 and 79 years now exceed the waist circumference threshold for central obesity. Central obesity is positively associated with coronary heart disease risk in women and men. It has been hypothesized that the sex differences in fat distribution may explain the sex difference in coronary heart disease risk. Even with the differences, at any given level of central obesity measured as waist circumference or waist to hip ratio, coronary artery disease rates are identical in men and women.
Iron-deficiency anemia is anemia caused by a lack of iron. Anemia is defined as a decrease in the number of red blood cells or the amount of hemoglobin in the blood. When onset is slow, symptoms are often vague, such as feeling tired, weak, short of breath, or having decreased ability to exercise. Anemia that comes on quickly often has more severe symptoms, including confusion, feeling like one is going to pass out or increased thirst. Anemia is typically significant before a person becomes noticeably pale. Children with iron deficiency anemia may have problems with growth and development. There may be additional symptoms depending on the underlying cause. Iron-deficiency anemia is caused by blood loss, insufficient dietary intake, or poor absorption of iron from food. Sources of blood loss can include heavy periods, childbirth, uterine fibroids, stomach ulcers, colon cancer, and urinary tract bleeding. Poor absorption of iron from food may occur as a result of an intestinal disorder such as inflammatory bowel disease or celiac disease, or surgery such as a gastric bypass. In the developing world, parasitic worms, malaria, and HIV/AIDS increase the risk of iron deficiency anemia. Diagnosis is confirmed by blood tests. Iron deficiency anemia can be prevented by eating a diet containing sufficient amounts of iron or by iron supplementation. Foods high in iron include meat, nuts, and foods made with iron-fortified flour.
The first docking of Soviet spacecraft was finally realized in January 1969 by the Soyuz 4 and Soyuz 5 missions. It was the first-ever docking of two crewed spacecraft, and the first transfer of crew from one space vehicle to another.

As of the mid-2020s retatrutide remains investigational and is not an approved medicine in the United States or the European Union. It has been supplied mainly to participants in clinical trials. Labels and availability can change, so regulatory listings should be checked directly.
Dual agonists act at the GLP-1 and GIP receptors, whereas retatrutide adds glucagon receptor activity on the same peptide. This third activity is the main structural and pharmacological distinction discussed in the literature. Direct head-to-head comparisons remain limited.
Reports usually give the study code LY3437943 alongside the name, the dose in milligrams, and the week at which an endpoint was measured. Percent body-weight change is the most frequent headline measure. Units and populations vary, so comparisons across reports require care.
